My first full-time position in flow cytometry. I became an operator of the flow sorter for clients from varied scientific backgrounds. A more technical role than I had previously held and working in a core facility. It allowed me to experience another aspect of grass roots scientific research.This involved helping clients with choice of antibody panels and fluorochromes to achieve the best results for sorting of cells to a specific population.I trained new clients to the facility in the use of flow cytometers for analysis. Ensuring that they were competent and more importantly confident to perform independent research.I introduced first-time clients to analysis software, in particular FlowJo.I wrote training manuals for clients to study themselves and ensure smooth running of the facility.I helped develop a new website for the facility, advising and deciding on content.(https://platforms.monash.edu/flowcore/)

I took this opportunity to expand my research experience into acquired immunology and specifically autoimmunity. Working with Dr. Patrick Bertolino on liver immunobiology. The lab had discovered an interaction between auto-reactive CD8 T cells and the liver. I was employed to investigate the molecular basis of this interaction and the resulting emperipolesis of the CD8 cells. This process resulted in the autoreactive T cells actually being consumed and destroyed by the liver. The project involved complex mouse-models of autoimmune disease, primary cell culture of hepatocytes and T cells, flow cytometry and protein-protein interaction assays.Hepatocyte entry leads to degradation of autoreactive CD8 T cells.
