Group of Prof. Achim KramerCo-supervisor: Prof. Matthias MelzigTitle: Regulation of CRY1 stability and its effect on the circadian period length.During my master thesis, I investigated the interdependence between CRY1 half-life and circadian periods by modulating CRY1 stability using C-terminal tags (e.g. SHIELD™, Pest-tag) and reintroducing these variants into CRY-deficient cells. I have used live-cell fluorescence imaging of circadian reporter cells and semi-automatic imaging analysis (ImageJ, CellProfiler) as well as western blot to assess protein half-life, and multi-day live-cell luminescence recording to measure the corresponding period phenotype.

A lab rotation in the chronobiology lab of Prof. Dr. Achim Kramer in fundamental molecular biology and biochemistry techniques. During this rotation, I mastered a broad spectrum of techniques, including the cultivation of primary and secondary cells, PCR, RNAi, protein and DNA gel-electrophoresis, and sub-cloning. I also become familiar with advanced methods, such as long-term live-cell bio-luminescence and fluorescence recording, and production of and transduction by lentiviral vectors.

During this time, I gained considerable experience in basic preparations of primary murine neurons and microglia, carried out mouse brain dissection, and mastered spinning disk confocal microscopy techniques and associated image analysis using ImageJ and Imaris software.

Melzig. During the project, I mastered the recently developed CRISPR/Cas9 genome editing toolbox, sapofection-based DNA/RNA delivery, PCR, and fluorescence-activated cell sorting (FACS).
