Led the biochemical characterization of chymosin aspartic proteases and supported the commercialization through the bioprocess development, scale-up and transfer to the production plant.

Led the Biochemistry Automation team in Palo Alto and drove the integration of capabilities with the other North American sub-teams. Led the wider rollout of the Protein Engineering data management systems and integration with the data management systems of other functions, namely those used by molecular biology and fermentation bioprocess. Continued to improve the cleaning performance and fitness of subtilisin proteases through protein engineering, while supporting the HTS campaigns for the protein engineering of various enzymes for Home and Personal Care, Biofuels, Food, and Animal Nutrition. Developed liquid-handling automation for viscous formulation excipients, insoluble solids, and complex mixtures down to 384-well scale for HTS of bioprocess conditions optimization. Optimized enzyme strains for C.E. (see below).

Led the small-scale performance evaluation of the enzyme strain responsible for hydrolyzing corn stover biomass for the generation of Cellulosic Ethanol for the DCE process. Developed a HTS screening system for the optimization of the enzyme composition to guide T. reesei strain construction, and a workflow to rapidly evaluate the performance of the strains co-expressing over 12 cellulases and hemicellalses.

Led the Home and Personal Care Applied Biochemistry team through the organizational integration between Danisco and DuPont and into the new Industrial Biosciences business division. Built a data management system for the capture, storage and analysis of HTS results from protein engineering campaigns through the customization and integration of third-party software tools. Continued to further improve and commercialize enzymes for Home and Personal Care applications through protein engineering, consumer relevant applications evaluation and joint customer innovation programs.

Led the Home and Personal Care Applied Biochemistry team in Palo Alto, building the local capabilities to assess enzyme performance in application relevant conditions. Integrated HPC applications research between Genencor and Danisco after the acquisition and merger. Developed and commercialized multiple enzyme products through protein engineering, application development, and joint customer innovation programs with key accounts which included cold-water proteases and amylases for cleaning in multiple formats and geographies.

Developed multiple enzyme technologies for the Home and Personal Care application. Characterized subtilisins, metalloproteases, and a novel perhydrolase for cleaning, then developed multiple orthogonal high-throughput screens for the protein engineering of each for improved performance and stability. Evaluated adjacent applications and markets where the use of peroxygen bleaching with the perhydrolase. Co-located at a large global consumer products customer to develop new products based on these new enzyme technologies. Additionally investigated the efficacy of subtilisins for skin care applications, catalases for fizzing and foaming in cleaning, and polyesterases for the removal of pills during fabric treatment and cleaning.

Wolfe Laboratory in Department of MicrobiologyMicrobiology and BiochemistryCharacterized phosphoenolpyruvate carboxykinase from Mycobacterium smegmatis to gain insight into the metabolic pathways leading to M. tuberculosis dormancy during pathogenesis. Cloned PCK from M. smegmatis into E. coli, expressed without purification tags, purified in native conformation, and characterized active enzyme. Published results in the Journal of Biological Chemistry.Investigated the oxidation of sulfur compounds by the photosynthetic green sulfur bacteria Chlorobium tepidum. Generated random knockout mutants using conjugative plasmids prepared in E. coli. and analyzed alterations in sulfide oxidation. Conducted growth and genetic manipulations of C. tepidum in a strict anaerobic environment. Supported work by a Colgate-Palmolive junior research grant.

Biophysics Lab of Dr. P. BuschInvestigated the effects of compounds on model human substrates, specializing in robotic assays and hair care at Henkel’s biophysics lab in Duesseldorf, Germany. The internship involved venturing into skin care applications; conducted two projects simultaneously. Measured the gloss and gas permeability of a model skin substrate, porcine skin, after treatment with experimental compounds. Devised an image analysis assay to measure skin gloss, a property which exaggerates wrinkled skin. Also devised a novel technique to prepare grafts of skin, for measuring gas permeability with a transcutaneous oxygen and carbon dioxide sensor.

Molecular Diagnostics Laboratory of Dr. Yuqi ZhaoMolecular Biology and Functional ExpressionInvestigated the effects of recombinant HIV Protease on the cytoskeleton of S. pombe by phenotypic evaluation of mutants. PCR amplified HIV protease genes from the cDNA isolated from infected patients and cloned into E. coli. Transferred plasmids to S. pombe resulting in phenotypic changes in cellular morphology, structures and metabolism. Presented results at the 1997 Seattle convention of the American Association for the Advancement of Science. As a side project, created a beta website for the MDL.
