About Eun-Jeong Won

During my research, I studied organ-specific targeting by lipid-based nanoparticles (LNPs, liposomes, microbubbles, and micelles). I have developed methods for encapsulating various biomaterials within these particles, including plasmid DNA, mRNA, saRNA (self-amplifying RNA), and proteins (CRISPR-Cas9 variants). Also, I have focused on evolving therapeutics by conjugating antibodies, plasma-membrane proteins, and DNA aptamers to the surfaces of these nanoparticles for targeted organ delivery. Building on these experiences, I am eager to broaden my research to include the study of various nanoparticles and polymers. Furthermore, I am interested in exploring the use of nanomaterials in both diagnostic technologies and therapeutic applications.* Available Experimental Skills *โ€ข Synthesis and analysis of lipid-based nanoparticles. (LNP, liposome, microbubble, micelle) 1) Materials suitable for encapsulation: NIR-dye (hydrophobic chemical compound), CRISPR-related proteins, pDNA, mRNA, saRNA. 2) Materials capable of surface binding: Antibodies, Membrane proteins, DNA aptamers. 3) Methods for nanoparticle synthesis: hand-mix, freezing-thawing, extrusion, microfluidics (ignite, T-junction). 4) Analysis of nanoparticles: Zeta-sizer, NTA, TEM, SEM.โ€ข Production and purification of exosomes derived from mammalian cells 1) Modified Mesenchymal stem cells method: liposomes loaded plasmid DNA, electroporation. 2) Purified MSC exosome method: size-extraction column.โ€ข In vitro study and analysis. 1) Cultured mammalian cells: cancer cells (pancreatic, breast, colon cancer etc.), immune cells, primary cells, mesenchymal stem cell (MSCs). 2) DNA & RNA study: gene cloning, PCR, QRT-PCR. 3) Synthesis of in vitro transcribed mRNA and saRNA. 4) Development of Cell based-DNA aptamer: tumor cells, proteins based-SELEX 5) Gene transfection. (transfection tools: LNP, lipofectamine, PEI, and electroporation) 6) Protein study: SDS-PAGE, WB, Immunoprecipitation, ELISA, Flow cytometry, and Confocal microscopy. 7) Protein expression in E.coli. & Protein purification. (affinity, size-extraction column, FPLC)โ€ข In vivo study and analysis. 1) Mouse modeling alopecia and xenograft tumor cells. 2) Administration routes : IP, SC, IM, IN, IT, and IV (basic). 3) Blood & body fluid collection techniques: orbital sinus, cardiac puncture and BALF. 4) Analysis of tissue protein levels : H&E staining, IHC, and IF. 5) Assessment of vaccine immunogenicity: antigen-specific IgG titers, ELISpot and flow cytometry analysis of splenocytes.
