Cloning and expressing fusion proteins in cells and mice using gene therapy to induce tolerance to autoantigens.More specifically, I designed a fusion protein that included both an endosomal/lysosomal targetting moiety and the immunodominant epitope from collagen type-II. When expressed in APCs, the collagen II epitope was presented on MHCII. When administered to mice (delivered using HIV-1-based lentiviral vectors) the incidence and severity of collagen-induced arthritis was reduced.
