About Elise Azar, Phd
It wasn’t my intention to be a PhD. I wanted to be an astrophysicist, a domain that didn’t exist in my country. Instead I did biochemistry, but on a day of march 2011 I read on the NASA magazine about the new potentials in a new scientific domain, the Biophysics field. May 2011 I applied to the only Biophysics master class in Paris, where only 12 students are taken and I was proudly accepted without even a prior meeting.This was the day my journey started!!!I'm known for my extreme optimism and commitment and I KNEW I am meant to BE a member of this discipline.Soon after I joined this master class I was captivated by the X-ray scattering and desperately wanted to pursue my studies in a PhD in this field. Persistence paid off!! I am now a multidisciplinary person (biochemist, biophysicist and PhD in physics).Being known for my entrepreneurial mindset, I created an internet service provider company (PnE ISP) in Lebanon at the age of 19 and I knew with the PhD it would be difficult to manage both so I sold it out. Being a manager and an owner of a small company with 4 employees at a young age made me very RESPONSIBLE and a PEOPLE PERSON. I became very FLEXIBLE and can MANAGE DIFFICULTIES. I learned how to be COURAGEOUS and take risks wisely and especially to BELIEVE in MYSELF and MY ideas.In 2015-2016 I worked on a doctoral mission during my PhD as a project manager at the SATT of Paris-Saclay university. I was in direct contact with the laboratories' directors with an objective of detecting innovative projects that would attract industrial collaborations. In order to… Show more The aim of this study was probing the interaction of membrane inclusions with their environment,(lipid or surfactant bilayers). Inserting nano-objects within the membrane has two typesof effects: a bilayer deformation which engenders a membrane-mediated interaction between the inclusions at the nanometer scale. And a second effect defined by local variations in the positional and orientational order parameter of the membrane constituents that occur at a “microscopic” scale. In order to study the membrane-meditated interaction between embedded inclusions we performed a systematic investigation on highly aligned and oriented lamellar phases using SAXS andvarying relevant parameters: membrane thickness, cholesterol content, temperature, degree of hydration, surfactant type, inclusions types and the inclusion density. The interaction potential was obtained from the structure factor of the scattered samples using the integral equations of the Ornstein-Zernicke with the Percus-Yevick closure. We found that the interaction potential decreases with the temperature and the inclusion content, and that the cholesterol plays a major role on the interaction since it changes the hydrophobic thickness of the membranes but its effect varies according to the type of inclusion used. Only at low hydration, the layers are closer and the inclusions are able to interact with their counterparts in neighboring layers.In order to study the influence of embedded peptides on the neighboring acyl chains we combined WAXS which gives access to the positional order between neighboring chains and “DROSS-NMR” which is sensitive to the orientational order of chain segments. Overall we have small modifications in the local order in terms of position and orientation when adding gramicidin (case of DMPC, DLPC and C12E4). Only in the case of DDAO, the gramicidin content seems to notably influence the acyl chains by decreasing their positional order and increasing their orientational order. Show less

For this purpose I studied the growth of this bacterium in culture media of different compositions by time-lapse microscopy and I estimated the growth rate and the size distribution of bacteria. It has been shown previously that the average size is an exponential function of growth rate. However, in a bacterial population sizes vary greatly from one individual to another. Size distributions corresponding to different growth rates were not previously characterized. According to my measurements in four different media, I found that, regardless of the growth rate, in other words, regardless of the culture medium, a universally-size distribution curve exists and is then adapted to a multiplicative factor depending on the medium. These bridges arguably are the most questionable structure component of PhK which for some are artifactually generated or altered by conventional sample preparation techniques for electron microcopy,The aim of my project was to calculate the structure of PhK in the nonactivated form at a higher resolution than that published and compare it with the activated form using the technique of single particle reconstruction from images obtained by Cryo-electron microscopy. To achieve high resolution, I had to work with hundreds of thousands of images selected from the projections of two different low resolution features one of the activated state consisting of two bridges (obtained by negative staining) and the other of the nonactivated state consisting of 4 bridges (obtained by cryo). I determined different classes in both cases with preferred orientations. Accordingly, calculated 3D models are all different. This suggests that the analysis technique and the references used also influence the outcome of the final structure. So in perspective, we must resume the selection of particles by checking the scripts and comparing with manual selection. Show less
