Our primary aim is to discern how matrix-targeted and cell-targeted C’ Dots interact with chondrocytes over a span of five days, with a hypothesis suggesting that cell-targeted C’ Dots will demonstrate enhanced internalization and prolonged retention compared to their untargeted or matrix-targeted counterparts.Our experimental methodology involves precise preparation of trimmed cylindrical cartilage plugs, followed by meticulous incubation with C’ Dots under controlled conditions. Advanced imaging techniques and sophisticated analytical tools are employed to meticulously track the retention dynamics of C’ Dots within chondrocytes throughout the experimental timeline.The findings from this research hold profound implications for targeted drug delivery strategies in cartilage regeneration and disease therapeutics. Our research focused on the engineering of therapeutic agents with the aim of specifically targeting osteoarthritis. Through our preliminary work, we successfully demonstrated the ability of C' Dots to penetrate the dense, avascular cartilage matrix and effectively reach resident chondrocytes within the tissue. Moreover, our functionalized C' Dots showcased a notable reduction in effective diffusivity, attributed to their reversible interactions with the matrix. In this project, my primary responsibility involved the engineering of C' Dots to bind with targets present in the cartilage matrix, thereby facilitating enhanced delivery of therapeutic agents. -

The original loader is machined (by a desktop mill) from solid piece of plastic (Delrin -acetal homopolymer), and metal bars are required to prevent bending from machining stress to ensureeven loading volume. The platform is composed of two parts: the omniphobicplates patterned with hydrophilic patches, and the loading device (loader) that deposits the liquid ofinterest onto the hydrophilic patches.The envisioned new variant will improve on a couple of fronts: 1) Enable variable “pick and use” loader configurations. 2) Inherit geometrical consistency, which strongly impacts the loading volume. 3) Metal rail + plastic insert design for customizability and reduction in cost of consumables. 4) Upscaling the manufacturing of such loaders. After the developed loader has been wellcharacterized, a proof-of-concept experiment will be performed. -
