My project in the Chapman Lab involves structural studies of Fatty Acid Amide Hydrolase (FAAH) isoforms in the legume, Medicago truncatula. Techniques I am using include recombinant protein expression in E. coli cultures, protein purification using metal affinity chromatography and size exclusion chromatography. Protein samples are verified using SDS polyacrylamide gel electrophoresis (SDSPAGE), and western blotting. Protein structure techniques include small angle X-ray scattering (SAXS) and macromolecular crystallography. Cryo-EM and small angle neutron scattering (SANS) experiments are also progressing.

In the Nam Lab, I was involved in studies focusing on the regulations of miRNA processing and biogenesis. Skills acquired include PCR, sub-cloning using the Gibson method (NEB), recombinant protein expression in E. coli, SF21, and Tni insect cell lines (Expression Systems). I also gained extensive knowledge and experience in recombinant protein purification using metal affinity, ion exchange, and size exclusion chromatography. My responsibilities also included in vitro RNA transcription and purification, and mouse colony maintenance. I am also experienced in electrophoretic mobility shift assays (EMSAs), western blots, and in vitro methylation.I also managed lab safety compliance including radiation safety, biohazard safety, and hazardous chemical safety.
