Scientific lead responsible for Purification (Downstream) Development at Catalent for the development team in Bloomington, Indiana. I oversee process design and characterization, and facilitate technical transfer to cGMP production.

My work as a scientist is focused on downstream process development. I plan and execute experiments supporting client projects before they transfer to manufacturing. I work in developing and optimizing processes as well as transfer of processes.

My work is focused on furthering our understanding of the GTPase Ras and its role in cancer. More specifically, I am interested in looking at differences in Ras complexes under various cellular conditions. Another focus of my work is to analyse the Ras-membrane interaction with more physiologically relevant membrane mimetics.

My work as a postdoc expanded on the work done as a graduate student studying the cytoskeletal protein EB1. My goal was to explore the differences in the EB proteins evolutionarily and determine if changes in the EB1 sequence could be attributed to duplication events or through specialization of EB function.

Characterized the EB1–actin interaction • Used a variety of biophysical techniques to determine the strength of the EB1-actin interaction in vitro• Mutated various amino acids of EB1 to determine which are responsible for interacting with actin• Transiently transfected EB1 in to culture cells to investigate the effect EB1 has on actin• Advised and mentored two new graduate students to become independent scientists • Managed several undergraduate students and mentored them through beginning and intermediate laboratory skills needed as technicians. Investigated of the mechanism of EB1-microtubule binding • Collaborated with a leading analytical ultracentrifugation expert to determine the affinity of EB1 dimerization using sedimentation velocity and sedimentation equilibrium centrifugation• Used computer simulation and biophysical techniques to determine the EB1-microtubule binding site• Determined that EB1 does not bind to the seam of the microtubule but rather to the microtubule body• Provided insight in to a new mechanism for EB1 regulation of microtubules in vivo

o Involved in setting up molecular assessment protocols for antibody therapeutic leado Analyzed antibody forced degradation samples using an improved digestion method for comprehensive coverage of chemical degradation products o Determined the sequence liability (focusing on Asn deamidation & Asp isomerization) of several antibodies using liquid chromatography mass spectrometry (LC-MS)o Executed tandem mass spectrometric experiments and data analysis to identify the site of chemical modificationso Initiated method development and implementation of analysis for released glycans from antibody and fusion proteinso Tested a new protocol for glycan release from antibody & fusion proteins and successfully identified the glycans released from the fusion protein

As a chemistry intern, I investigated the process of radiator failure through engine coolant- radiator interactions.More specifically, I determined that the interactions of the engine coolant and the new coating on the surface of the radiator were causing radiator failure through production of gelatinous material. I also reproduced in vitro the precipitate formation and analyzed the precipitate through various techniques including visual inspection, FTIR, and chemical analysis.
