Project Goals: To elucidate the epigenetic mechanisms regulating the expression of Th17-specific genes including IL-; 22, and determine the mechanisms by which Th17 cells suppress expression the Th1 effector cytokine IFN-gIdentify regulatory elements within genomic DNA that may modulate expression of Th17-specific genes using computational predictions, high-throughput, assays and targeted approaches.Characterize putative regulatory elements and determine whether they are enhancers, boundary elements/enhancer blockers or silencers. Assess the 3D chromatin looping structure at the Il17a/f gene locus in various helper T lymphocytes lineages.Determine which cis-regulatory elements are brought in proximity with Th17 gene promoters when the locus is being expressed.

Studied alleviation of specific cell-cycle constraints involved in negative regulation of cell cycle, namely, the cyclin-dependent kinase inhibitors p27kip1 and p18ink4c. Assessed the effects of p27kip1 and p18ink4c loss on in vitro anergy induction, proliferation and cytokine production by CD4 T cells. Examined effector function of T cells deficient in p27kip1 and p18ink4c in vivo, using a murine model of cardiac allograft rejection.Evaluated time to graft rejection in wild-type mice or p27kip1 and p18ink4c knockout mice receiving anti-rejection therapies that are known to slow or halt T cell responses.
