Dissertation Title: Maternal Programming of BehaviorWorked on two projects involving non-genetic maternal transmission mechanisms that influence offspring brain development and behavior. The first involves TNF-deficient mothers who bear offspring with abnormally increased hippocampal proliferation and, ultimately, augmented spatial memory and contextual fear response, as a result of reduced TNF-regulated chemokines in her milk. This work identified a programming function of maternal TNF on hippocampal development and memory. The second project involves a serotonin 1A receptor (5-HT1AR) deficient mother and the cellular and behavioral consequences of that deficiency on her offspring and even her grand-offspring. In humans, chronic stress and depression are associated with reduced 5-HT1AR levels. The offspring of 5-HT1AR deficient mice exhibit increased anxiety (avoidance and hypoactivity) and stress reactivity, a phenotype indicative of psychiatric disease. The gestational effects of the maternal receptor deficit are not transmitted genetically, but rather, via parallel and segregated mechanisms and are not transmitted altogether. The traits of anxiety, avoidance and hypoactivity, are transmitted somatically, while increased stress reactivity is transmitted through epigenetic signatures on germ-cell DNA. This research addresses a central problem in the epidemiology and diagnosis of psychiatric disorders; particularly, how their symptoms are propagated across generations and why they may present differently in individuals, even those in the same family tree. For example, family members can have various combinations of anxiety, depression, bipolar disease and schizophrenia symptoms, complicating diagnosis and treatment. This work may facilitate the development of more patient-specific and combination approaches for the treatment of psychiatric conditions.
