At the University of Florida, I gained comprehensive experience in natural product isolation and enzyme characterization in bacteria, ranging from molecular gene cloning and expression through structural determination. This includes skills such as designing and implementing genetic engineering experiments for bacterial expression, microbial culturing including both solid and liquid fermentations, chemical extraction and purification, and small molecule analysis via HPLC, LC-MS, UV-Vis, GC-MS, and NMR. Additionally, I have extensive experience in protein characterization and purification techniques such as SDS-PAGE and FPLC (ÄKTA Pure). This is reflected in my publication “Structure-guided product determination of the bacterial type II diterpene synthase Tpn2,” in which I designed and tested multiple protein mutants to determine the importance of specific residues during biosynthesis.
