Postdoctoral Research Associate
My postdoctoral research included the continuation and optimization of topics from my PhD work, including the chromatographical and spectroscopic analysis of various proteins and the structural characterization by X-ray diffractometry. Furthermore, I established an electrochemical method for the quantification of protein midpoint potentials in the workgroup and was responsible for the re-validation of existing methods. In addition, I wrote and monitored SOPs for lab-internal methods and supervised the transfer of knowledge that I obtained throughout my research stay to other PhD students of the group.

My work as a researcher has ranged from cloning and gene editing through cell cultivation and protein purification to the characterization of purified protein samples:- Gene editing by site-directed mutagenesis and lambda-Red recombination- Homologous and heterologous protein production by cell cultivation in various growth media and conditions in small to medium (4 mL to 28 L) scale, including flask- and fermenter cell growth- I gained a lot of experience in protein purification techniques and strategies including IMAC, SEC and IEC using FPLC- and HPLC-systems.- I validated protein sample composition and purity by BN-, SDS- and SAR-PAGE.- Protein characterization by spectroscopic methods: UV/vis-, fluorescence-, IR- and EPR- spectroscopy- I analyzed enzyme kinetics: steady-state substrate conversion and ROS formation; pre-steady-state kinetics by stopped-flow and microsecond freeze-hyperquenching- The thermal stability of proteins was analyzed by ThermoFAD, Thermofluor, nanoDSF and SEC-TS- I co-developed and validated a novel fluorescence-based Thermofluor approach for the characterization of protein stability in collaboration with Dr. Jo Hoeser.- I investigated the influence of sample storage conditions on protein stability in freeze-/thaw- and lyophilization- experiments.- Protein-small molecule interactions were determined kinetically and by MST or ITC.- I solved several new protein variants and protein-inhibitor structures by X-ray crystallographyI taught these techniques to supervised students, my fellow PhD students and postdoctoral colleagues, especially in regard to protein purification and spectroscopic as well as thermostability characterization. Additionally, I concepted, validated and implemented lab courses, I co-wrote publication manuscripts and gained experience in presenting scientific posters and presentations in the lab or at national and international conferences.
Student Researcher And Tutor At The Institute Of Organic Chemistry And Biochemistry
During my studies of chemistry, I worked as a student researcher and tutor in the context of the chemical lab course for medical students and students of the biology teacher training at the Institute of Organic Chemistry and Biochemistry of the Albert-Ludwigs-University in Freiburg. My tasks as a student assistant included:1. preparation and follow-up processing of chemicals and lab equipment in the practical course rooms2. introduction to aspects of laboratory safety (e.g. fire-extinguishing exercises)3. demonstration of experiments4. guiding students in experimental work5. assistance with the correction of the written examination and6. answering questions about the theory and the experiments of the individual days of practical training.My tasks as a tutor included:1. answering questions about the theory of the individual course days and linking the material of lectures and seminars to the topics of the practical course.2. guiding the students in solving the examination tasks. The students learned how to apply knowledge from the lectures and seminars to specific questions.Furthermore, as a simple learning aid, I have compiled summaries of organic and inorganic theory in the form of fact sheets that are still in use today.

I had the great opportunity to work as a student researcher in the workgroup of Prof. Dr. Oliver Einsle. I was supervised by Dr. Anja Pomowski and collaborated with Prof. Dr. Martha Sosa-Torres (Facultad de química, Universidad Nacional Autónoma de México, Mexico City, Mexico) in a project focused on studies of protein purification and crystallization of a PQQ-dependent alcohol dehydrogenase (QADH) from Gluconacetobacter diazoazotrophicus. Through this collaboration, I gained great skills in chromatographic protein purification techniques and strategies as well as in protein crystallization and X-ray diffractometry experiments.
