• Provided data processing and analysis support to HTS scientists, including the use of appropriate software and the interpretation of results. • Provided first-line support in diagnosing data capture and processing errors occurring in the platform's automation workflow. • Gathered user functional requirements and worked with software engineers in implementing in-house data processing solutions.

• Developed and executed high-throughput screening assays for various biological systems and assay technologies. Examples include: modulation of bacterial pathogenicity using T. pyriformis/K. pneumoniae predator/prey relationship to mimic macrophage/bacteria interactions; biochemical and enzyme screens using time-resolved fluorescence; cell-based screens using luciferase reporter genes, for both agonistic and antagonistic modes of action.• Programmed lab benchtop and robotically integrated instrument systems to automate assays. • Developed assay miniaturization capabilities, including optimization of assay development, liquid handling, and data analysis. • Used design-of-experiment (DOE) techniques to optimize assays. • Identified high-throughput data analysis bottlenecks and created scripts to speed data flow and reduce time spent manually manipulating data sets. • Evaluated newly developed in-house HTS data analysis package, providing scientific end-user perspective to developers. • Designed and set up a new screening laboratory to move microorganism assays (C. elegans, yeast, Drosophila, and cells) into high throughput screening formats. This entailed - evaluation and purchase of appropriate robotic technologies, including liquid handling and detection; - institution of a compound storage management system; - development of assays and execution of high-throughput screens; and - implementation of data management software to automatically extract data from screens and perform preliminary analyses. • Developed and performed a screening assay to find compounds which affect orphan nematode nuclear receptors in a yeast-based reporter gene system. • Created databases in FileMaker Pro which cataloged compounds and microorganism strains used for screening and correlated primary screening results across several projects.

ENZYMOLOGY/HTS GROUP. Development and execution of various high-throughput screening assays, such as:Optimized a kinase assay for automated compound screening using AlphaScreen bead-based proximity readout. Created cell lines with secreted alkaline phosphatase reporter construct and used those cells to screen for compounds which interfere with the NF-kappa-B signaling pathway. Used cell-based high-throughput screening techniques to identify lead proprietary compounds which selectively kill tumor cells during hypoxia. Created and maintained databases in FileMaker Pro which cataloged in-house inventory of proprietary and purchased compound libraries used for high-throughput screening. Researched and designed a new robotic screening system for the HTS lab.
