Investigating tumor microenvironment characteristics to enhance the efficacy of cellular immunotherapies and myeloid-cell modulators to treat multiple types of brain cancer, including glioblastoma multiforme (GBM), IDH-mutant astrocytoma, oligodendroglioma, and anaplastic meningioma.

Block, MD, PhD (Div. of Medical Oncology, Dept. of Immunology)Developed novel peptide, neoantigen, dendritic cell, & HLA-based tumor vaccines for melanoma, including spearheading experiments in preclinical mouse models and assisting phase I clinical development efforts. Led immunologic and serologic biomarker analysis for a phase I tumor vaccine trial for melanoma (NCT02320305), and served as lead immunologist and bioinformatician for a phase II melanoma combination immunotherapy trial (NCT03554083). Assisted in the development and validation of a novel multiplexed immunofluorescence (MxIF) immune monitoring platform to identify immune cells in patient solid tumor and lymph node samples. Created a unique mouse model of antigen-specific tolerance in order to investigate tolerance-breaking tumor vaccines, and explored a novel, adoptive B cell therapy strategy for the treatment of epithelial ovarian cancer in mouse models.Scientific skills used: mouse procedures, viral transduction, multicolor flow cytometry, antigen-specific T cell identification, ELISPOT, CyTOF, R.

Investigated novel adjuvants (e.g. TLR3, TLR4, STING agonists) across peptide vaccine formulations to develop an ideal platform for tumor neoantigen peptide vaccination. Deployed protocols for high-sensitivity isolation and single-cell RNA-sequencing of immune cells from murine skin and lymphoid tissues. Evaluated multiple preclinical neoantigen vaccine candidates in humanized mouse models of metastatic melanoma.Scientific skills used: mouse procedures, lymphoid & non-lymphoid tissue analysis, multicolor flow cytometry, light microscopy, single-cell RNA-sequencing, ELISPOT.

Paid research internship at Aduro Biotech, a clinical-stage company developing novel immunotherapies such as Listeria-based vaccines and STING agonists for multiple cancer types. Performed research under Dr. Kelsey Gauthier in the Department of Immunology. Completed multiple DNA sequencing projects on human and animal tissue samples to characterize STING alleles that determine responsiveness to subsequent agonistic cancer immunotherapy. Designed and executed multiple cell-based assays to validate the physiological & pharmacological effects of first-in-class STING agonists and other CDNs. Analyzed patient samples to assist the clinical validation of Listeria-based tumor vaccines in a phase II study for lung cancer (NCT01675765). Performed in vitro toxicology testing to assess purity and safety of multiple drug candidates. Presented research at department meetings and contributed data to IND & regulatory findings, assisting the pre-clinical development and testing of CDNs as STING pathway activators against cancer.Scientific skills used: mammalian tissue culture, immunological assays, experimental design, microscopy, PCR, Sanger sequence analysis, gel electrophoresis, flow cytometry, Luminex.

Harvested organs from mice immunized with a novel antigen, isolated cells for the production of B cell hybridomas, evaluated growth of antibody-secreting cells to select clones for expansion, and quantified antibody production via assays such as ELISA and IP/SDS-PAGE/Western Blot.Additionally, used DNA manipulation techniques such as PCR, gel electrophoresis, restriction enzyme digestion, plasmid cloning, and Sanger sequencing to characterize the unique V(D)J recombinations of multiple B cell samples.
