Postdoctoral Fellow
Independently designed and fabricated a prototype microfluidic device for the immobilization and microdissection of individual cells. Using micro-3D printing (µ3DP), a laser-based rapid prototyping method, optimized the microfluidic device specifications in four fabrication cycles. With the optimized prototype, designed and fabricated an 8-fold higher-throughput device, the fission yeast lifespan microdissector (FYLM), using DraftSight and eGlade (Python-based design software). Developed a FYLM protocol to conduct high-throughput parallelized cell-aging studies of the fission yeast Schizosaccharomyces pombe. The FYLM provided a stable environment for the automated, full-lifespan observation of the yeast cells for >100 hours and >50 generations, with minimal human interaction. Developed MATLAB functions for analyzing raw data, to reveal changes in cell morphology as cells age and elucidate mechanisms of cell aging.
Postdoctoral Fellow
Developed a method for fabricating millimeter-scale hydrogel structures with micron-scale features using multiphoton lithography (a 3D printing technique). The method required writing code in the ImageJ macro language to convert 3D virtual objects into a timed sequence of linear masks, and writing LabView code to coordinate the mechanical and optical components used to fabricate the lithographic structures.Wrote MATLAB code to model interaction between a spherical force spectroscopy probe and a surface of finite thickness. The model allows better understanding of the relative importance of parameters used in studies on the mechanical properties of micron-scale hydrogels.
Graduate Research Assistant
Work focused on using femto-second pulsed lasers to fabricate micron-scale scaffolds from biomaterials for use in tissue engineering applications. As part of Ph.D. dissertation research, developed a material allowing the fabrication of unsupported micron-scale hydrogels, enabling the first fabrication of such structures with biological polymers.Responsible for design and execution of experiments studying the mechanical and functional properties of the hydrogel scaffolds. Wrote MATLAB code to determine the Young’s modulus of micron-scale hydrogels using raw force spectroscopy data.Significant contributor to both an NSF Major Research Instrumentation grant application and an NIH Exploratory/Developmental Research (R21) grant application.Mentored 5 undergraduate students, helping them to develop, fund and execute research plans.
