Profile bio

About Erwin Pannecoucke

Understanding how things work at a fundamental level, and using that knowledge to tackle problems. This is, in a nutshell, what triggers my interests in both my professional and personal life.Core competencies:---------------------------------7 years of experience in a top-level academic research environment, with close collaborations with various biotechnology companies on the development of their next-generation products. Expert in protein production and purification, biophysical characterization (X-ray crystallography, isothermal titration calorimetry, multi-angle laser-light scattering, biolayer interferometry) and protein engineering (creative site-directed mutagenesis and cloning). Experienced user of third-generation synchrotron light sources of EMBL Hamburg (Germany), ESRF Grenoble (France), SOLEIL (France) and SLS (Switzerland).Extensive expertise in protein visualization and in silico manipulation tools (PyMOL, Coot), crystallographic analysis and modeling (XDS, CCP4 Package, Phenix, Buster), GitHub (personal usage and in collaborations) and molecular cloning (SnapGene).Expert user of Microsoft Windows (including WSL), Linux-based operating systems (Ubuntu, CentOS, Raspbian) and high-performance computing (in-house CPU/GPU workstations, Vlaams Supercomputer Centrum / HPC UGent).Enthusiastic autodidact in Python and Bash programming, home automation (Home Assistant) and Android development (OS and UI level). For fun, I try to deploy a self-hosted and privacy-friendly alternative for every cloud-based service that I'm using, mastering Docker in due course. In general, I'm skilled in quickly understanding the grammar underlying programming languages, and how code translates into a specific program behavior. Team player by nature; scout from the age of 6, scout leader for 6 years and until recently, almost 10 years accountant of it's underlying non-profit organization.

Listed skills include Isothermal Titration Calorimetry, X Ray Protein Crystallography, Linux, Molecular Cloning, and 11 others.

Senior Scientist Discovery

Current

Gent En Omgeving

As a biochemical and biophysical expert at argenx, I support early Discovery projects by providing technical expertise and troubleshooting support across various stages, from cloning over protein production and characterization, to antibody engineering. With a strong foundation in structure-function relationships, I enjoy tackling complex problems and exploring innovative techniques to uncover new insights.I'm passionate about staying up-to-date with the latest technologies, diving into the fundamental principles of novel instruments, but also deploying the latest containers for an in silico characterization to enhance our understanding of biochemical interactions. Finally, I've led the development and implementation of our current LIMS/ELN system, and continue to drive its refinement and strategic expansion. A key aspect of that role is help defining the data strategy for Discovery and explore how we can leverage that data, combined with computational capabilities, to augment our wet lab processes.

R&D Scientist

Zwijnaarde

Mar 2021 - Mar 2023

Post-Doctoral Fellow

Gent

In addition to finalizing running projects, I closely collaborate with 3 industrial partners. Using our multidisciplinary platform of biophysical techniques, protein X-ray crystallography and cellular tools, we aim to get deeper insights on products either early in the developmental pipeline, either already in Phase 3 of clinical testing. Regardless the state of the product, we proactively use the gathered knowledge to provide suggestions on how to improve and validate the behavior of their next-generation drugs.

Mar 2019 - Feb 2021

Phd Student At Ghent University - Vib Center For Inflammation Research

Zwijnaarde, Belgium

My PhD research was mainly focused on understanding how single point-mutations in the extracellular region of the transmembrane receptor FLT3, identified as driver mutations in AML primary samples, could exert such dramatic effect on the intracellular activity of the receptor. Gathering such insights required a challenging multidisciplinary approach, combining several biophysical techniques with protein X-ray crystallography and cellular studies. Mastering techniques as isothermal titration calorimetry (ITC), biolayer interferometry (BLI), X-ray crystallography, flow cytometry and statistic is what was finally required to provide unique insights in the molecular mechanistics , architectural principles and structure-function relationships underlying the extracellular receptor activation.
