• Optimized the upstream production of extracellular vesicles (EVs) derived from megakaryocytes. This work aimed at both improving both differentiation to and proliferation of megakaryocytes, as well as the production of EVs, and involved studies into media components, temperature, donor input, impurity removal, and incubation duration, to name a few.• Performed downstream purification operations such as tangential flow filtration (TFF), ultracentrifugation, density gradient ultracentrifugation, and antibody-based bead isolation.• Loaded EVs with functional cargo and characterized the loaded products with regard to particle size and concentration, encapsulation efficiency, and copy number.• Characterized populations of EVs determining the key features which distinguish them from other particles present in the process and those which delineate their loaded forms. This work used NanoFlow (Flow Cytometry with appropriate filter), NanoFCM (flow cytometer sold by NanoFCM), zetasizer, and Nanoparticle Tracking Analysis (NTA).

• Determined the effectiveness of new unit operations and process alterations at pilot plant scale for an upstream process producing an erythroid lineage from hematopoietic stem cells. Some examples of this work were: the addition of a media exchange using TFF (Tangential flow filtration), use of exhaust condensers, and alterations made to media recipes.• Prepared and characterized solutions used in upstream and downstream processes across multiple scales (0.2L – 400L). In this regard, experimental work characterized the impact of freeze thaw cycles, degradation with aging, and helped determine the appropriate tests to measure solution quality.• Created procedures for process stages and characterization techniques.• Performed data analysis using Spotfire and Pi datalink.
