During my time as an intern at AVA, I was tasked to identify Curvularia species in Axonopus Compressus using morphological and molecular detection. Techniques I acquired include isolation of Curvularia, extraction of DNA and Polymerase Chain Reaction (PCR) of ITS and GPD14 gene which were known to amplify DNA of Curvularia species. Subsequently, DNAs were sequenced and blast against NCBI database for the identification of Curvularia species.Besides being involved in a project, I assisted in the daily laboratory diagnostic of plant pests identification. It includes several laboratory techniques such as identification of different fungi, preparation of culture media, sterile techniques as well as bacteria/fungal isolation from soil samples. Nevertheless, observations of pathogenicity test (koch’s postulates) and other laboratory skills greatly enhance my knowledge.
