The main focus of this internship was to assess the role of alternative polyadenylation (APA) of mRNAs in cancer. I was responsible for the cloning of both the coding sequence and the UTR of two candidate genes, Next we studied the functional role of APA with respect to these candidate genes on mRNA stability, protein expression/activity and localization, by using qPCR, Western blot, and luciferase assays and confocal microscopy. Furthermore, I have also learned to design shRNAs.

The goal of this internship was to study the potential angiostatic interaction between endothelial cells and platelets via galectin-1. This interaction was studied using the following techniques: isolation of primary human umbilical vein cells (HUVECs), migration assay, proliferation assay, platelet isolation and activation determined by flowcytometry, and aggregation assay.

During this period I was responsible for culturing fibroblast-like synoviocytes of patient material, PBMC isolation, cutting tissue using a cryostat, and different (multi) immunohistochemical or immunofluorescence stainings on frozen and paraffin embedded tissue. The stainings were performed either manually or by the usage of an autostainer and quantified using semiquantitative analysis (SQA), digital image analysis (DIA), or confocal microscopy.

In this internship the role of the chemorepellent/receptor pair Slit and Roundabout in migration and metastasis formation of neuroblastoma cells was studied. I applied a knockdown strategy (both transient as lentiviral) and verified knockdown using flow cytometry and W-blot. Next I characterized these cells for migration using a transwell assay and confocal microscopy. Furthermore, we have also confirmed the interaction between Slit and roundabout homologues by immunoprecipitation
