My thesis demonstrates that the cellomics field can be revolutionized by integration of novel label–free imaging technologies, i.e. spontaneous Raman microscopy and lens-free imaging (LFI). I worked on applications of these imaging techniques in cancer research and focused on two metastatic key players that are heavily researched on the single cell level, i.e. circulating tumour cells (CTCs) and cell migration.My PhD work demonstrated the first application of LFI for live-cell imaging. Individual cancer cells were time-lapse imaged while migrating in microstructures in order to assess their migration phenotype. A wealth of information about the cells' migration behaviour could be extracted from their paths. Also metastasized cancer cells showed higher migration speed and directionality when subjected to more confined microenvironments. Live-cell imaging was achieved with LFI using coherent laser illumination, increasing the quality of the individual holograms. This enables the combination of large field of view, single cell morphology resolution and fast acquisition times necessary for live-cell imaging.I used spontaneous Raman spectroscopy to distinguish individual cancer and blood cells, showcasing the importance of data analysis and background removal for single cell classification to be based on the biology of the cell. Also, the classification results were linked to differences in molecular cell content using principal component analysis. This indicates that Raman spectroscopy might be suited for CTC identification in blood samples, enabling better disease/therapy follow-up and faster cancer detection. Show less
