1) Led CRISPR-based gene editing in prostate cancer, including the design and development of a CRISPR construct, the validation of the gene editing (disruption) in cancer cell culture using PCR, RT-qPCR, and Western blotting, the confirmation of the loss-of-function studies by mirroring siRNA and compound induced inhibition assay. 2) Guiding the in vivo functional investigation by using a mouse Xenograft model.3) Collaborated on a team that investigated the lipid metabolism in isolated mouse primary hepatocytes after the gene delivery through injection of AAV expressing liver-specific Cre into mice, and determined the molecular functions of protein of interest (POI) in cholesterol efflux. 4) Major research worker in the project on Liver Function and Metabolism: primary hepatocyte isolation, Mitochondrial and microsomal subcellular fractionation, and Seahorse running.5) Carried out fatty acid metabolism and cell proliferation assay in inhibitor-induced and CRISPR-induced cell culture.6) Performed lipid isolation, TLC assay, cholesterol turnover assay and efflux assay.7) Published 2 papers, trained 3 students, worked with 1 RO1 and submitted 1 new RO1 grant.

1) Led the investigations of protein oxidation and acetylation in sepsis-induced acute inflammatory disease.2) Led the regulations of histone deacetylase and the inhibitors in bioenergy consumption and immuno-metabolism.3) Led the protein acetylation modulation in macrophage cell and animal models of sepsis combining high-fat-induced chronic inflammation.4) Published 10 papers, trained 3 students, completed 1 RO1, one new RO1 got supported.5) Organized and managed lab running, controlled the project progress.

1) Participated in gain- and loss-of-functions in the obesity-induced inflammation by compound treatments, as well as the delivery of recombinant constructs (lipid-based transfection of plasmid, electroporation, and lentivirus or AAV transduction) expressing the cDNA or shRNA, highlighted the importance of the vagus nerve in obesity development, and the epigenetic manipulation during macrophage (primary cells and cell line) M1/M2 polarization.2) Published 6 papers, trained 1 student, and one RO1 got supported.
