1) Led CRISPR-based gene editing in prostate cancer, including the design and development of a CRISPR construct, the validation of the gene editing (disruption) in cancer cell culture using PCR, RT-qPCR, and Western blotting, the confirmation of the loss-of-function studies by mirroring siRNA and compound induced inhibition assay. 2) Guiding the in vivo functional investigation by using a mouse Xenograft model.3) Collaborated on a team that investigated the lipid metabolism in isolated mouse primary hepatocytes after the gene delivery through injection of AAV expressing liver-specific Cre into mice, and determined the molecular functions of protein of interest (POI) in cholesterol efflux. 4) Major research worker in the project on Liver Function and Metabolism: primary hepatocyte isolation, Mitochondrial and microsomal subcellular fractionation, and Seahorse running.5) Carried out fatty acid metabolism and cell proliferation assay in inhibitor-induced and CRISPR-induced cell culture.6) Performed lipid isolation, TLC assay, cholesterol turnover assay and efflux assay.7) Published 2 papers, trained 3 students, worked with 1 RO1 and submitted 1 new RO1 grant.
