Buss TraineeUniversity Of Zurich Jul 2009 - Aug 2009Zurich Area, SwitzerlandETH/UZH BUSS: international Biology Undergraduate Summer School.During the two months I spent at the Institut für Molekularbiologie (UZH), Zurich, my main research objective was the study of some interesting aspects of epigenetic regulation, with special attention to chromatin modification. Chromatin modification is a very important mechanism of gene expression regulation. One class of enzymes involved in this process is the family of SET domain containing histone methyltransferase (HMTase) enzymes, which are highly conserved in evolution from yeast to man. The yeast Set1 complex methylates histon 3 on its lysine 4 (H3K4) and this modification is a landmark for actively transcribed chromatin. In our current research we identified a H3K4 mimicking domain in the Set1/c corepressor protein Nrm1. This domain was shown to directly mediate the interaction of Nrm1 with two important cellular complexes: both Set1c and Cdc20/APC complex. This work aimed to identify proteins also containing an H3K4 mimicking domain by in silico screening of the yeast proteome. Five candidates proteins were then tested for interaction with Cdc20. We could show that the Yjr142w, a protein of unknown function was able bind Cdc20 suggesting a cell cycle dependant regulation of Yjr142w by the APC. Bsc Thesis TraineeUniversity Of Parma Apr 2009 - Jun 2009Parma Area, ItalyThe main purpose of the research I carried out at the Biochemistry Department (UNIPR), Parma, was the cloning and subsequent assembly of a Magnaporthe grisea gene, whose product appears to be homologous to protein family members CVNH. The work assigned to me was, first, the extraction of cellular mRNA M. grisea, then its reverse-transcribed into cDNA, as well as assembly and amplification via PCR. This was followed by the cloning of the sequence obtained in an E. coli expression vector, the transformation of a strain suitable for its production, as well as a subsequent phase of selection of the correct recombinant.
