I am driven by a passionate curiosity for the natural world and I see my life as an enduring quest for knowledge. My current work centers on bringing together cancer genomics and metabolism, to develop novel molecular targeted therapeutics. We have pioneered a novel approach to precision oncology, collateral lethality, which utilizes passenger deletions of housekeeping genes as points of metabolic vulnerabilities. Our most advanced iteration of this strategy consists of targeting tumors with ENO1 homozygous deletions (occur in GBM, HCC, CHOL) by inhibitors of the redundant paralogue, ENO2. We generate a small molecule ENO2 inhibitor, HEX with pro-drugs of HEX showing potent pre-clinical activity and good tolerance in NHP. At the same time, I see passenger deleted genes as natural experiments that can inform on key aspects of tumor biology, including immune system/tumor interactions.

Postdoctoral FellowDana-Farber Cancer Institute Oct 2007 - Dec 2011Boston, Ma, UsMy post-doctoral training was in the lab of Ronald DePinho on Mayer 4 at Dana Farber Cancer Institute. While I initially joined the lab to work on the biology of aging, this incredible environment in cancer hospital setting stimulated my interest in cancer metabolism and therapeutics. During the first few years in the DePinho lab, I studied the role of Foxo transcription factors in aging and neurodegeneration in close collaboration with JiHye Paik. Our key finding was that neuron-specific knockout of Foxo1 results in a dramatic acceleration of aging-related axonal tract degeneration. Other collaborations with Mariela Jaskeliof and Ergun Sahin, concerned the role of telomerase in aging. -
Graduate Student ResearcherGraduate School Of Biomedical Sciences At The University Of Texas Health Science Center San Antonio 2002 - 2007San Antonio, Texas, UsMy graduate research concerned the mechanism of reaction oxygen species (Superoxide) production by the mitochondrial electron transport chain and its pathophysiological implications for Aging and Neurodegeneration. Highlights from my work during this time, include the finding that Superoxide produced at the Qo site of mitochondrial Complex III can reach both sides of the inner mitochondrial membrane and hence exert oxidative damage in both the mitochondrial matrix and the cytosol. The physiological importance of cytosolic superoxide is highlighted by the dramatic acceleration of aging-related sarcopenia that we observe in Sod1-/- mice. -
