About Florian Noack

During my PhD and Postdoc I gained deep theoretical and practical experience in a wide variety of next gene sequencing technologies including (sc)RNA-seq, (sc)ATAC-seq, ChIP-seq, WGB-seq, MethylHiC, HiC, NOME-seq and developed a new multiomic method. Additionally, I am familiar with bioinformatic pipelines to analyse the obtained data (R, Signac, Seurat, ArchR, Bismark, DeepTools, cell-ranger, NGS.plot, bedtools, MACS, etc. ). Besides that, I am experienced in a broad range of (molecular-)biological methods including mammalian in-vitro (human cancer- and mouse embryonic stem cell lines) as well as in-vivo (mouse surgery, in-utero electroporation) model systems, CRISPR-Cas9 based (epi-)genome editing, ImmunoFACS, molecular cloning etc.

Identification and validation of gene regulatory elements for the next generation of gene therapies. During my Postdoc, I developed a novel multiomic next generation sequencing approach which allow to measure simultaneously gene expression, chromatin accessibility, DNA methylation and 3D chromatin structure. Together with commercial (single cell-) multiomic methods and an integrative data analysis strategy, I was able to identify gene regulatory elements during mouse and human brain development. I successfully completed my diploma thesis with the title “Analysis of gene expression and epigenetic regulation in the dentate gyrus of mice living in an enriched environment” in the laboratory of Prof. Dr. Gerd Kempermann. During my project, I compared the expression of various neurogenesis related genes in the dentate gyrus of mice housed in standard or environmental enrichment cages. Based on these data, I chose candidate genes to analyze the DNA methylation pattern of their promotor regions. For this purpose, I introduced a method called bisulfite sequencing which allows mapping methylated cytosine of a specific region with a single base resolution.Key techniques:• surgery and dissection of mice• behavioural test (morris water maze)• bisulfite amplicon sequencing• establishment of real time RT PCR assays
