Group "Bone and Ligament Regeneration", Head: Darja Marolt, PhDresearch focus: cargo analysis of bone marrow mesenchymal stem cellsMy aim is to improve bone regeneration with a cell-free extracellular vesicle-based approach. Therefore, I conditioned bone marrow mesenchymal stem cells that were almost 100% confluent. In order to purify the extracellular vesicles, I performed an ultracentrifugation step following FACS and NTA analysis. The goal of my master thesis is to analyse the cargo of the bone marrow mesenchymal stem cells.

In this course the students can get exciting insights into everyday's working life of researchers. Furthermore, the students gain advanced knowledge in microbiology and genetic engineering by presentations and experiments organised by the tutors.

My main achievement was the successful testing of several small to medium scale liquid chromatography runs. This included buffer optimization, column packing with different matrixes, programming of the chromatograph, minimization of time consumption and comparison to recent literature. I also succeeded to establish an internal contact databank in MS access that helped to build an overview of the internal network with contact details and product affiliation if applicable. Furthermore, I helped with general secretarial work.

Kolling Institute of medical ResearchGroup "Cardiovascular Regeneration", Head: Prof. Gemma Figtree, Dr. Carmine Gentileresearch focus: 3D in vitro models to study cardiac angiogenesisDuring my Bachelor Internship I was in charge of creating three-dimensional vascularised heart organoids out of stem cells. After creating the models, I was responsible to bioprint them within an alginate-based hydrogel where I assessed cardiac angiogenesis enhancement with VEGF, cell viability and organoid migration and fusion with immunohistochemistry staining for different cell markers. Furthermore, I immunohistochemically processed and analysed paraffin embedded mouse heart sections. I presented the work at two different international conferences in Australia, and two in Austria, secured a travel grant for Annual Scientific Meeting of the Australian Vascular Biology Society and claimed three different international and national prices for my work.
