During this period my research was focused on the design, synthesis via MCRs and preliminary biological evaluation of new small molecules targeting the IL-17A homodimer.Since IL-17A homodimer and its receptor play a major role in many inflammatory diseases, inhibiting their action could be a valid and alternative approach for patients who are not responsive to the widely used TNF inhibitors. The application of synthetic macrocycles and small molecules to challenging protein-protein interactions targets represents an important emerging area, with potential implications for the future balance of effort between biological and small molecules drug discovery. Multicomponent reactions (MCRs) require less reagents, save time and effort, and allow a fast probe of a chemical hypothesis. In this way very complex target molecules can be obtained by combining simple, often commercially available, educts. Exploiting reactions such as Ugi, Ugi-Tetrazole, Passerini and Passerini-Tetrazole reactions, I was able to build a library of nearly 60 IL-17A inhibitors. The most potent showed a nanomolar activity.

During my master's thesis my project was focused on the design, synthesis, and preliminary biological evaluation of 3-cyclopropyl-4-phenoxy-1H-pyrazole derivatives as small molecular ligands of Receptor for Advanced Glycation Endproducts (RAGE).
