I worked part-time as a research assistant at the Diabetes, Obesity, and Metabolism Institute at the Icahn School of Medicine at Mt. Sinai in New York City. I worked in Dr. Andrew F. Stewart's lab, whose research focuses on the use of dual-specificity tyrosine phosphorylation-regulated kinase 1A (DRYK1A) inhibitors such as harmine (which his group discovered) as a means of inducing beta cell differentiation and proliferation. Sinai in New York City. I worked in Dr. Andrew F. Stewart's lab, whose research focuses on the use of dual-specificity tyrosine phosphorylation-regulated kinase 1A (DRYK1A) inhibitors such as harmine (which his group discovered) as a means of inducing beta cell differentiation and proliferation. My junior mentor, Dr. Courtney Ackeifi, published her thesis discussing the use of the glucagon-like protein 1 receptor (GLP-1 receptor) on beta cells and the signal-transduction pathway that is involved as means of promoting beta cell replication in conjunction with harmine, other DRYK1A inhibitors, and different agonists for the GLP-1 receptor. The research I have been involved with directly focused on using harmine and GLP-1 in HepG2 cells to observe the effects of each drug on different proteins involved in the cell cycle pathway, such as p21, p27, cyclinA and CREB/pCREB. As part of the first cohort of medical students at Stanford University under the NIDDK Medical Student Research Program, I was able to partake in research regarding beta cell proliferation and targeting a known beta-cell proliferation drug, GNF-4877, to the rat R7T1 beta cell using a zinc chelator, and comparing the data to zinc-deficient equivalent cells that were achieved via zinc-transporter 8 knockout (ZnT8 KO). I received training on how to properly grow and divide cells, as well as how to plate cells properly. I also learned how to freeze and unthaw cells. In addition, I learned how transfect these cells with DNA plasmids, as well as create more DNA plasmids using bacterial transformation with competent cells. In a similar manner, I learned how to grow competent cells. In addition, I learned how to treat the beta cells with different drugs in a dose-dilution series, as well as how to properly lyse the cells and transfer the lysed material into a plate for reading. I learned how to use two different plate readers to gather data, and how to properly plot this data onto dose response curves using Excel and Prism GraphPad. I also learned how to calculate the relative efficiency using the Ec50s from the dose response curves for both cell types with each drug treatment. Learning all of this information will further my career as it will allow me to design and conduct similar experiments on my own, and has increased my interest in conducting research in diabetes as part of my physician career.
