I am the Risk Manager for the primary IVD software for new assay on QIAGEN's flagship PCR platform. My main role at cobra was the purification of biopharmaceuticals, most notably DNA. I was regularly involved in the purification of over 100mg of research grade plasmid DNA. Additionally, I've also played a significant role in the development of the purification method for DNA produced by multiple displacement amplification techniques. This included optimising the small scale process and development of this procedure for scale-up to GMP mass-production, with the target of producing 1g of DNA without the need of a bioreactor. Furthermore, my analysis of routine procedures resulted in the identification of several areas for streamlining and simplifying regular tasks. This included replacing a semi-qualitative method with a more reproducible quantitative method, plus the complete removal of an entire processing step from a separate procedure.
Postgraduate Researcher
I was responsible for the expression and purification of human nuclear receptors using affinity, ion exchange and gel filtration chromatography. This then allowed me to characterise the heme ligation by UV-visible and EPR spectroscopy, and the DNA-binding and transcriptional activity using functional assays.In addition to this I was involved in the development of a new method for the determination of reduction potentials, responsible for solving the structure of a protein from the electron density, writing research papers for publication and regularly involved in teaching. I gained approx. 100 hours experience demonstrating to undergraduates in the teaching labs and supervised and assisted new PhD students, master students and undergraduate project students working in the lab. Furthermore, I was heavily involved in running and maintaining the lab, including the ordering of consumables and liaising with external companies when equipment needed to be fix or replaced.
