About Giancarlo Vecchio

Giancarlo Vecchio has been Director of the Department of Molecular and Cellular Biology and Pathology at the University of Naples Federico II.He has been Associate Editor of Cancer Research. Is Associate Editor of Oncology Research, International Journal of Oncology, Tumori, Thyroid Research and of The Faculty of Medicine 1000. He has devoted much of his scientific life to Oncology. His work has been initially addressed to retroviruses, a category of viruses strictly related to cancer. Since research on retroviruses has led to the discovery of oncogenes, he has addressed his interest to this category of genes, one of the most important targets of genetic alterations of tumor cells. He showed that retroviral and cellular oncogenes (mainly ras genes) had an oncogenic potential toward epithelial cells.The work in his lab has been centered upon the role exerted by oncogenes in thyroid oncogenesis. A novel oncogene has been identified and cloned in his lab in1987.The gene (RET/PTC) is activated by chromosomal rearrangements, represents the first human oncogene isolated in Italy and is responsible of a high percentage of papillary thyroid cancers. His research group has shown that the gene rearrangement responsible for the activation of RET/PTC is a very frequent event in thyroid papillary carcinomas of children of the Chernobyl area. In his lab it has also been shown that point mutations of the RET gene are responsible of both familial and sporadic medullary type of thyroid carcinoma. These types of new findings, concerning human oncogenes in thyroid cancer, have brought fundamental insights into the molecular pathogenesis of human neoplasias and are now leading to new therapeutic treatments for these diseases. In 1995, for his contributions to cancer research, Giancarlo Vecchio has received the Fogarty Scholar in Residence award at the National Institutes of Health (U.S.A.).He is currently Director of the Istituto Superiore di Oncologia.

    My current research interest is concerned with the molecular mechanisms of metastasis. I am involved at the present time with research on a lysosomal enzyme, the alpha-L- fucosidase (FUCA-1), a glycosidase whose action consists in removing the monosaccharide L-fucose from glycoprotein moyeties. The expression of the enzyme is reduced in aggressive metastasizing cancers of the breast and colon, whereas it is conserved in less aggressive and non metastasizing cancers. I am at the present evaluating the expression and the role of this enzyme in aggressive metastasizing thyroid cancers such as anaplastic thyroid carcinomas versus less aggressive thyroid cancers such as papillary thyroid carcinomas.

    During the period in which I was Director of the Department I have continued to be involved in research concerning the molecular pathogenesis of human thyroid cancer. I have been involved in research concerning the activity of bovine pancreatic ribonuclease and of a recombinant human ribonuclease, which both have a dimeric structure, on human thyroid cancer cell lines. We have demonstrated that these ribonucleases exert a strong inhibitory action upon human and rat thyroid cancer cell lines,whereas they exert almost no inhibitory activity against normal rat and human thyroid cell lines. Monomeric human pancreatic ribonuclease has no inhibitory activity against either normal or tumorigenic cell lines. The activity of the dimeric ribonucleases is at variance with what was observed with common anticancer drugs, such as doxorubicin, which exert a more potent inhibitory effect against normal than against tumorigenic cell lines. Thus dimeric ribonucleases seem to carry a specific inhibitory action against tumor cells and can be considered as potentially useful drugs for cancer therapy.

During this period, in collaboration with several younger researchers, we have demonstrated that the rearrangement of the RET/PTC oncogene occurred frequently in papillary thyroid cancers of young children of the Chernobyl area, where the frequency of this tumor in children was higly increased.

    During my research period at the Institute for Molecular Virology at St.Louis University Medical School, I have been working in collaboration with Dr. Nobuo Tsuchida, Dr. Saumya Bhaduri and Dr. S. We were able to demonstrate that polyribosomes from cells stably infected and producing the murine Moloney leukemia-sarcoma retrovirus contain the virus specific messenger RNA.
