Development of CosMx Spatial Molecular Imager (SMI)○ Developed new reagents for SMI Protein and RNA assay to reduce background and improve signal. ○ Developed SMI morphology markers for accurate cell segmentation by screening antibody clones and writing custom image processing scripts to quantify improvement in staining quality and segmentation. ○ Developed image processing pipeline for protein SMI Protein assay by quantifying various methods for background removal to improve signal to noise ratio (SNR). ○ Collaborated with biostats and engineering to quantify improvements to cell typing and recommended improvements to commercial software. ○ Analyzed data and contributed results to internal and external publications and posters. Development of the GeoMx Spatial Proteogenomic assay – Nanostring’s first multiomic workflow that enables high-plex spatial profiling and quantification of RNA and Protein on the same FFPE section. ○ Point person for data analysis in R.○ Confirmed assay performance and compared data to external RNASeq dataset from the Cancer Cell Line Encyclopedia. ○ Contributed results to a technical white paper and one scientific publication.
