The aim of my PhD project has been to decipher the pathological cellular and molecular mechanisms underlying hereditary nephrotic syndromes, in particular of forms associating neurological disorders to the kidney disease. To achieve this goal, my project mainly focused on the establishment of in vitro tools to obtain meaningful “genetic disease in a dish” models. I thus differentiated kidney organoids and neural stem cells from patient-derived induced pluripotent stem cells (iPSC). This new iPSC-based approach allowed to highlight cellular phenotypes linked to hereditary diseases on relevant cell types and in a patient-specific context.

The aim of my M2 interniship has been to characterize a recently indentified population of putative prostate luminal progenitor cells. Based on their androgen-independence these cells are excellent candidate to be involved in prostate cancer recurrence after escape to anti-androgen therapy. I validated the transcriptomic signature of these progenitor cells, showing that they represent distinct cellular entity in the prostatic epithelium. I identified specific markers to establish their relevance in various contexts of mouse and human prostate tumorigenesis, starting to gain insight the molecular mechanisms that govern their proliferation and their actual oncogenic potency.
