Biochemistry Lab Researcher - Senior Honors Thesis project. Summary: Prepared and analyzed an altered anticoagulant protein (HCII) to measure the effect of the changes on its role in blood clottingProject Details: We altered the N-terminal domain of a well-known blood protein called Heparin Cofactor II (HCII) to test a model of its proteolytic function. Preliminary data using an arterial mouse model demonstrated that said model seems to be critical to the function of HCII in vivo.*Utilized digest enzyme to delete specific acidic domains in the N-terminal region of Heparin Cofactor II (HCII) which is an anticoagulant glycoprotein that inhibits thrombosis by a covalent 1:1 interaction with the serine protease thrombin. *Used a photochemical agent to damage the carotid artery HCII-deficient mice and inserted altered protein to observe the effect of digested HCII variants on the rate of thrombosis.

Developmental Biology/Genetics Lab AssistantSummary: Studied control mechanisms in a zebrafish overgrowth mutant. Produced genetically mixed fish to assess whether the overgrowth gene acts at the location of overgrowth or throughout the entire system. ---Highlights:* Utilized plasmids to create mosaic fish and then florescence microscopy to trace the GFP reporter in order to address the origins of cells comprising the regenerated structures in the caudal fin of the zebrafish. * Dissected global and local growth control mechanisms in the zebrafish mutant rapunzel. Generated mosaic fish using heterogenic blastula transplants to address whether the rapunzel protein acts through a cell autonomous or non-cell autonomous mechanism. * Performed control experiments on a transgenic line of zebrafish to observe the effect of a heat shock promoted gene in conjunction with the mutation of interest. *Using camera microscopy and NIH Image software: Compared vasculature and regeneration in the mutant rapunzel to that of wild type in the zebrafish model. Measured the quantity and quality of regeneration in the Rapunzel zebrafish mutant as compared to that of wildtype.
