Transcriptome characterization and biomarker discovery in Atlantic salmon affected by Amoebic Gill Disease. This project aims to characterise the acute phase of Atlantic salmon immune response in AGD challenge experiments by means of transcriptomic and proteomic approaches. Salmon strains displaying varying levels of resistance to AGD will be challenged with N. perurans and tissue samples will be collected over a period of time throughout progression of infection. Differentially expressed genes will be identified by means of Next Generation Sequencing (NGS) technology (RNA-seq). Further characterisation of selected genes will be carried out by means of standard PCR and Real Time quantitative PCR (RT qPCR) allowing further investigation of transcriptome acute phase immune response between salmon strains displaying varying levels of resistance to N. perurans and the role of protein translation rate with tolerance or resistance to N. perurans infection. This project will generate knowledge on the immune response of farmed Atlantic and will allow the identification of potential biomarkers to be implemented in the future development of targeted therapies and disease mitigation strategies

Testing a wide range of both pharmaceuticals & biopharmaceuticals and medical devices in a GMP laboratory. Duties include:Water testing, Endotoxin testing, Bioburden testing, Growth Promotion tests, QC checks on media used in lab, Antimicrobial effectiveness testing (PETs), Disinfectant testing, Environmental Monitoring and Identification of micro organisms, cytotoxicity testing and Microbial Limit Testing. All tests performed to EP or USP guidelines.Also responsible for ordering consumables (media etc) and general stock for day-to-day running of the laboratory. While applying cGLP to all laboratory activities including stock control.Interpreting sampling and monitoring data, applying statistical and analytical techniques, to determine control and trending.

Responsible for all raw material testing in lab using methods such as pH, appearance,solubility,Thin Layer Chromatography (TLC),Karl Fischer, Potentiometric titration, FT-IR and UV.Preparation of reagents, mobile phases and samples used on Waters Alliance HPLC systemsAnalysis of solid and liquid dose pharmaceuticals.Calibration and operation of analytical instruments including Karl Fisher, conductivity meter, pH meters, densitometer and analytical balances. Participating in out of spec investigations and LIR's.Undertake change controls, updating test methods, product specifications, and quality assurance reports using current pharmacopoeias (EP,USP and BP).Correcting and reviewing of other peoples lab reports

Responsible for performing sample and solution preparations, carrying out and accurately documenting analyses performed using detailed methods following SOP and cGMP requirements. Responsible for evaluating and reporting analytical data in accordance with approved protocols and specifications. Involved with project status reporting to project leader.

During this placement I completed a project titled ‘’Validation of a Loop Mediated Isothermal Amplification Assay for the Detection of Group B Streptococcus in pregnant women’’. I also designed a poster demonstrating my work on this assay for presentation at the European Congress of Clinical Microbiology and Infectious Diseases 2014.
