We outlicense s50 technology to partners developing new therapeutic candidates to create next-generation therapies, particularly in the fields of RNA and pDNA drugs. s50 is differentiated in part by its novel size and structure that enables extrahepatic targeting as well as cell uptake via non-degradative caveolae pathways, similar to viruses such as SV-40.Key accomplishments:• Discovered, patented and developed stabilized micelle, co-crystallization nanocapsule chemistry for cell and organ-specific non-viral delivery of nucleic acids, contrast and small molecules via IV, subcutaneous, inhalation, and passive topical routes.• Early adopter and ongoing optimizer of 3D cell culture methods, crucial to proper understanding of endosome-avoiding, lipid-raft trafficking s50 capsules.• Successfully scaled chemistry and process from 125 ug to 144 mg to support studies inrodents, rabbits, weanlings pigs, and companion felines.• Led GMP scaleup of recombinant protein for proprietary s50 tumor-targeting ligand.• Advanced analytical methods and solid-state characterization of novel solid dosage form toward GMP scaleup.• Developed novel cold isotopic method for in vivo biodistribution based on NAA. • Demonstrated tumor-targeted checkpoint inhibition for lead compound in oncology.• Expanded and further tested s50 capsule chemistry to include mRNA, RNP and other unique CRISPR constructs, protein therapeutic and microscopy reagent cargos.• Recruited, trained and led interdisciplinary laboratory team of staff and consultants.• Authored or co-authored more than 30 journal articles and conference posters, inventor on 15 patent applications, and Principal Investigator or key collaborator on 19 awarded SBIRs and other federal and state grants and contracts.
