As a Post-Doctoral researcher at National Cancer Institute, NIH, I am exploring the mechanism of gene regulation (transcription) at single molecule level. I have developed a Single Molecule Tracking (SMT) method to measure the dynamics of the transcription activators at specific gene promoters in the yeast Saccharomyces cerevisiae. I use computational modeling and simulation approaches to understand the biophysical parameters measured from SMT. This work has been selected for the platform presentations at the Cold Spring Harbor Laboratory, New York in 2018 and the American Society for Cell Biology meeting, San Francisco in 2016. This work lead to two high impact publications, six conference presentations and two invited talks. The method developed here (SMT) has a high potential in future research to measure the fast dynamics of the bio-molecules in vivo.

My Doctoral research focused on the mechanism of chromosome dynamics (cell division) and genome instability using a eukaryotic model system S. cerevisiae. Meiosis is a specialized cell division process required for the production of the gametes. Combining Cell, Molecular, Biochemical and Imaging approaches, I explored the structural and functional differences in the kinetochore, a multi-protein complex that connects centromeres to microtubules, between mitosis and meiosis. I also explored the mechanisms of reductional chromosome segregation during meiosis, known as protection of the centromeric cohesin and sister kinetochore monoorientation. This work led to ~8 international peer reviewed publications, 3 international and 3 national conferences. This work provides a fundamental understanding about the meiotic cell division, a process required for gametogenesis, failure of which leads to anueploidy, still birth, developmental disorders or cancers.
