    • Leukemia, a cancer of the blood, has a poor prognosis with current treatments having high incidence of systemic toxicity and development of resistance. Defined as immature white blood stem cells that uncontrollably reproduce, STAT5 plays a major role in the progression and maintenance of this cancer, inducing the continual production of these leukemic stem cells. With STAT5 being validated as an important diagnostic marker in the governance of stem cell fate, targeted therapeutics designed to block this protein’s activity may provide for an effective strategy in counteracting the overproduction of white blood cells. The Gunning Group was the first to report a rationally designed therapeutic, with great specificity for the STAT5 protein. They identified AC-3-019 as the most potent STAT5 inhibitor reported to date. In this proposal, we aim to use medicinal chemistry to rationally optimize AC-3-019 to identify a hit-to-lead compound, suitable for advanced preclinical trials. The long-term objective is to identify an analog of AC-3-019 with increased half-life and in vivo stability for clinical application.• I assisted in the synthesis and characterization of a library of second-generation inhibitors at the University of Toronto Mississauga (Department of Chemical and Physical Sciences). Inhibitors were evaluated for biological activity via fluorescence polarization assays.
