In charge of gene therapy vector (AAV) production

- Successfully established a scalable baculovirus system for recombinant adeno-associated virus (rAAV) vector production at 25-liter scale.- Purified large amounts of rAAV vectors using column chromatography for preclinical studies of hemophilia and Gaucher gene therapies.- Cloned more than two hundreds of different plasmid constructs for the company’s various gene therapy projects.

- In charge of projects for creating recombinant adenovirus that carries human interferon b gene and establising stable E-1 expressing cell line to complement E1-deleted adenovirus.- Completed the project of generating recombinant adenovirus carrying AAV rep/cap genes for AAV production and filed patent.- Completed the project of generating recombinant baculovirus carrying adenovirus helper genes and AAV rep/cap genes for AAV production and filed patent.- Design and constructed AAV vector, AAV packaging, and adenovirus helper plasmids - Established a 293-based cell line with rAAV-GFP stably integrated.- Constructed a total of 120 different plasmids for the company’s different research projects.- Screened mouse liver cDNA library and fished out 4 different genes for hyperlipidemia project.

- Generated a series of constructs expressing differently modified hexon proteins to study the host immune response to hexon proteins. - Generated adenovirus vectors containing transposase and transposon signals for integration studies using the cre-lox recombination system. - Constructed lots of plasmids ranging from 3 kb to 30 kb. - Generated retroviruses expressing different transgenes.
