I am fascinated by biology, and endeavor to contribute to our understanding of the genomes and genetics of model organisms. I believe that pieces of information can be put together to solve global issues in human health. In particular, I am interested in host-pathogen interaction, genomics, antibiotics and drug discovery for discovery and production of compounds of therapeutic use. Last but not least, I am dedicated to education as it is key to the development of our society.

Discovery and development of novel antimicrobialsIn the last several decades, more and more multi-drug resistance bacteria have been detected, but very few new antibiotics have entered the clinic and very few research dollars have been spent on development of next-generation antimicrobial agents. Recent report of the World Health Organization shows that global emergence of antibiotic resistant bacteria is posing a serious threat to the world public health. The more antibiotic resistant strains emerge, the fewer our drug choices and more expensive the drugs are. Modern medical advances such as surgery and transplants may face threats of infection. Antimicrobial research is thus pressingly demanded. A number of bacterial pathogens require the type III secretion system (T3SS) to grow inside host organisms. Fortunately, T3SS is largely absent in beneficial and non-pathogenic bacteria. This makes the T3SS an ideal target for novel antimicrobial agents. I am developing a high throughput screen to identify a suite of small molecules that can inhibit T3SS for use as new therapeutic candidates.

Virulence regulation in the plan pathogen Pseudomonas syringae pv.tomato DC3000The type III secretion system, required for suppression of plant defense and pathogenicity, is directly regulated by a sigma factor HrpL. HrpL regulon has long been a subject of several studies. RpoN is the sigma factor responsible for hrpL transcription. RpoN regulon is of great interest because RpoN is a global regulator for bacterial growth, survival and response to different environmental changes and presumably it coordinates the expression of other pathogenesis-related genes in parallel with those downstream of HrpL.Characterization of the HrpL regulon:• Globally mapping HrpL binding sites (using ChIP-Seq) and transcription of downstream genes (using RNA-Seq) across the DC3000.• Studying the roles of HrpL regulated genes in pathogenicity through mutational analysis.• Trapping promoters upstream of green fluorescence proteinreporter to search for functional promoters• Characterizing a novel effector protein.Characterization of the RpoN regulon:• Finding RpoN regulated genes using ChIP-Seq and RNA-Seq analysis.• Finding differentially expressed genes as pathogenicity related candidates
