Characterized activities of T7 RNA Polymerase and RNAseH enzymes that drive the oscillatory circuit.■ Developed in vitro assays for CRISPR-associated protein 9 (cas9) and cas9-like enzymes. The assay has been used for high-throughput screening of potent inhibitors of cas9 enzyme activity with success. Multiple patents have been filed for this work.■ Developed a conditional switch for in vitro activation of cas9 cutting. -
Postdoctoral ResearcherBoston University Mar 2011 - Aug 2013Boston, Ma, Us■ Developed a novel Next Generation Sequencing (NGS) based technique to map genomic DNA strand breaks created by gamma radiation in yeast (S. Cerevisiae) and human cells.■ Did basic NGS data analysis to predict functional open regions of chromatin and to obtain in vivo footprints of DNA-binding proteins.■ Used the NGS assay to map strand breaks and probe chromatin structure in frozen cell samples without the invasive thawing step.■ Developed a novel library preparation protocol where genomic DNA extraction and the first adapter ligation is done inside agarose plugs. -
