I validated novel mRNA vaccine antigen designs through in vitro potency assays (primarily flow cytometry and western blots) using mRNA I generated via cloning and IVT and transfected into adherent cell culture plates. I also produced recombinant proteins for many of these antigens in suspension cell culture flasks at up to 1L scale and purified them using size-exclusion chromatography for characterization with bio-layer interferometry and thermal shift assays to determine binding kinetics and melting temperature.By collaborating frequently with colleagues working on other teams, I applied creative solutions from different projects to my own work and vice versa, whether that be for assay development and refinement or data analysis. I also became very comfortable providing training on instruments and techniques to other researchers, and frequently either generated mRNA and protein for other teams to use and analyze or ran assays for other teams using mRNA and protein they provided from their own work.My other main responsibilities included adherent and suspension cell culture maintenance for multiple mammalian cell lines, mRNA transfection using both in-house formulations and commercial transfection reagents, cloning workflows using modern PCR, Gibson, and Golden Gate assembly techniques, plasmid preparation for storage and sequencing, sequence analysis and validation, data processing and visualization, and meticulous maintenance of electronic lab notebooks.
