LRRK2 - Leucine Rich Repeat Kinase 2, is a very important gene in human body. Mutations in this gene will cause Parkinson's disease. We selected the I1371V LRRK2 mutation for our study since it is an uncommon mutation that is only prevalent in east India and has not garnered much attention from researchers. In this study, three distinct plasmids containing the genes encoded for wild type lrrk2, I1371V and GS2019S mutant variant were transfected into SHSY-5Y cells. The cells were treated with GW5074, a GTpase inhibitor, and MLi-2, a kinase inhibitor, in order to determine their, effect on the levels of lrrk2 expression and substrate (pRab8a and pRab10) hyperphosphorylation, by immunocytochemistry and immunophenotyping respectively The MTT assay is used to determine their cytotoxic effects on SHSY-5Y cells transfected with LRRK2 mutant variants (GS and IV). This study indicates that there is no significant change in LRRK2 expression upon 24 hours of treatment with MLi-2 and GW5074. From this study we can infer that MLi-2 was found to be a better LRRK2 kinase activity suppressor in GS2019S mutation and GW5074 in case of I1371V.
