    Application of an existing pharmacological neonatal therapeutic in Cystic Fibrosis. Laboratory based skills; collection and transport of human biological samples from the Belfast City Hospital Regional Adult Cystic Fibrosis Unit and subsequent processing of sputum samples. Separation of mucus plugs from any salivary contamination in the sputum samples, processing by either 0.1% DTT (Dithiothreitol) or for use directly in experiments with surfactant. Samples were evaluated using an "in-house" fluidimeter to test for changes in the fluidity of the mucus or for chloroform-methanol extraction of lipids using Folch's method (1957). Lipids then analyzed by Thin Layer Chromotography. Development of frog model of mucociliary clearance using aerosolized channel blockers in order to simulate the CF situation for study of surfactant on various aspects of mucus clearance. In vitro skills; culture of immortalised A549, Cystic Fibrosis and Human bronchial epithelial cell lines. Use of said CF and HBE cell lines to test anti inflammatory potential of naturally derived surfactants with or without alpha tocopherol supplementation by IL-6 and IL-8 ELISA and MTT assay, respectively. Use of wound repair model-with A459 cell line only-to investigate effectiveness of surfactant ± alpha tocopherol supplementation by wound area analysis and IL-6 and IL-8 ELISA. Culture methods included: maintenance, division, handling and storage techniques and manipulation of experimental conditions.
