My thesis project focused on understanding the molecular mechanisms of transcriptional regulation in osteocytes (bone cells) and their response to the vitamin D hormone.- Molecularly dissected in vitro (mammalian cells) and in vivo (mouse) target enhancer regions using CRISPR/Cas9 mediated genome editing (transfection, flow cytometry, high-throughput testing), molecular cloning (restriction enzyme/ligase, Gibson Assembly, mutagenesis), reporter transfection, bacterial artificial chromosome… Show more My thesis project focused on understanding the molecular mechanisms of transcriptional regulation in osteocytes (bone cells) and their response to the vitamin D hormone.- Molecularly dissected in vitro (mammalian cells) and in vivo (mouse) target enhancer regions using CRISPR/Cas9 mediated genome editing (transfection, flow cytometry, high-throughput testing), molecular cloning (restriction enzyme/ligase, Gibson Assembly, mutagenesis), reporter transfection, bacterial artificial chromosome recombineering in E. Coli and stable cell line generation, protein analysis (Western blot), and qRT-PCR - Analyzed enhancer deletion effects in vivo (general mouse husbandry, genotyping, mouse tissue harvesting, RNA isolation, expression analysis with TaqMan® qRT-PCR)- High-throughput small molecule screening in cell culture systems (screening assay design, execution, QC analysis)- Investigated genome-wide transcriptional and epigenetic changes that occur during osteocyte differentiation (sterile mammalian tissue culture) using high-throughput sequencing techniques (ChIP-sequencing and RNA-sequencing) - Bioinformatics processing and analysis of genome-wide data sets - Recorded and portrayed scientific results through documenting, writing, and designing figures to effectively communicate in meetings and publications- Managed several projects concurrently while collaborating with colleagues - Mentored two undergraduate students in their thesis research projectsAdditional courses:- Wisconsin Entrepreneurial Bootcamp (2013) - a one-week intensive training program in technology entrepreneurship for graduate students in the sciences- Strategic Management of Innovation (2015) - a semester long class designed to help graduate students gain tools, concepts and information about how to generate value from technology.
