Current• Molecular gene editing projects:o Assumed a leadership role with 3-4 direct reports, while actively contributing as a team member. Provided guidance by teaching specialized techniques, organizing tasks, and overseeing the collective workflow, fostering a collaborative and efficient working environment.o Designed CAR-T construct for cloning in iPSC-derived T cells to achieve either gene Knock-out or Knock-in. o Performed Lygase-independent cloning of mentioned constructs using Infusion cloning, including multiple fragments and stepwise cloning methods. o Designed guide RNAs for multiple targets in-silico & screened for their efficiency in multiple cell lines.o Generated CRISPR knockouts in primary T cells and cell lines using Cas9, Cas12a, and MAD7.o Generated stable cell lines by viral transfection methods.o Designed and conducted PCR and sequencing-based TCR re-arrangement verification for in-house generated iPSCs (Recipient of SPOT Award)o Streamlined processes for Sendai viral remnant check, Mycoplasma contamination check by PCR detection methods. • Immuno-oncology projects: o Optimized immuno-oncology assays for large molecule screening and assessed numerous bi-specific and tri-specific antibodies.o Conducted binding and cytotoxicity assays with mentioned antibodies using Primary T cells against cancer cell lines in co-culture.o Developed and executed spheroid formation protocol for in vitro assays (utilizing GFP-tagged cell lines and Incucyte assay) and in vivo tumor generation (comparing both Matrigel & Cultrex) in mice to evaluate test antibody efficacy. o Conducted TCR-T reporter assay with client-generated TCR-T cell lines, targeting NFAT using luciferase and bioluminescence methods.
