About Hwoi Chan Kwon

As a biophysics PhD candidate at UC Davis, I have conducted research on the molecular mechanisms of ion channel function and regulation. My current project focuses on the slow gating of CLC-0—a prototype chloride channel in the CLC family—and its modulation by intracellular protons. Using patch-clamp electrophysiology and fluorescence microscopy, I've discovered and characterized a novel inactivation mechanism of CLC-0.I'm also a teaching assistant for UC Davis biochemistry courses and teach the fundamentals of protein structure, function, and metabolism, in addition to the concepts behind biochemical techniques.

    Generated novel mutations using recombinant DNA technology by designing primer oligo for site-directed mutagenesis, performing polymerase chain reaction, transforming E.Coli using heat-shock and electroporation, amplifying plasmid vector, mini prepping plasmid, and quantifying nucleic acid using UV-Vis spectroscopy.Experienced in culturing, harvesting, and prepping various expression systems ranging from adherent mammalian cell lines, such as HEK293 and CHO to X. laevis oocytes. Performed lipid-based transient transfection for heterologous expression in the mammalian cell line within the guidelines of a biosafety level 2 (BSL-2) environment; performed manipulation and injection of X. oocytes with mRNA load using the nanoliter injection system.Identified and quantified heterologous expression and activity of recombinant protein using wide-field fluorescence, total internal reflection fluorescence (TIRF), and fluorometry.Designed and performed patch-clamp electrophysiology using pCLAMP to generate thousands of high-quality recording traces using voltage and current clamp protocols, leading to the discovery of a novel gating mechanism for a CLC channel.Performed and analyzed two-electrode voltage clamp (TEVC) and single-channel recordings in quantifying macroscopic and microscopic properties of ion channels. Analyzed electrophysiology data using regression and statistical methods on Clampfit, excel, and Originlab (analogous to Graphed).Wrote technical papers and reviews and collaborated on original research, as demonstrated by first and second author publications.
