Performed research at the intersection of materials science, synthetic/systems biology, and computational biophysics to understand how non-muscle cells generate the requisite forces to drive fundamental mechanical processes, including cell division, motility, cell polarization, cancer metastasis, and tissue morphogenesis. I engineered biomimetic models of the actin cytoskeleton in vitro & performed computational modeling to investigate the sensitivity & scaling of contractile forces in disordered biopolymer networks. Further, I studied the influence of myosin active stresses on the architecture, dynamics, and stability of the actin cytoskeleton, as well as its role in driving local and global cellular shape transformations in non-muscle cells. The underlying physical principles that endow the actin cytoskeleton with its unique material properties can be applied to engineer active (i.e. non-equilibrium) materials, complex networks, and bio-inspired systems capable of self-assembly, internal stress generation, shape change, self-healing, and molecular transport for both medical and engineering applications.• Designed & conducted in vitro biochemistry & confocal microscopy experiments and analyzed the resulting imaging data sets• Engineered light-responsive biological materials that deform locally in regions exposed to visible light • Quantified biopolymer alignment & correlated motion in self-assembling liquid crystals & disordered networks and implemented an automated image analysis pipeline to extract these metrics from large microscopy data sets• Performed agent-based computational modeling of non-equilibrium protein networks and complex biological systems and developed an automated data analysis pipeline to efficiently analyze the large simulation data sets• Led research projects, managed interdisciplinary teams, and communicated research results to diverse audiences
Teaching Fellow: Biotransport And Kinetics
Biotransport and Kinetics | BME Department | Instructor: Professor Mark Saltzman• Led weekly discussion sections (~15 undergraduates)• Created lesson plans, section materials, problem sets, test questions, and solutions• Graded homework assignments and exams• Organized review sessions, and responded to student questions
Graduate Research Assistant
• Developed image analysis algorithms & automated workflows to efficiently quantify the material properties, structure, and protein dynamics from images of the disordered actin cytoskeleton in Xenopus laevis oocytes & in vitro biomimetic models
Undergraduate Research Assistant
• Designed and constructed targeting vectors to facilitate incorporation of custom engineered gene circuits into the genome of induced pluripotent stem cells (iPSC) via the CRISPR/Cas9 system• Developed intuitive software & lab protocols for creating custom CRISPR/Cas9 donor vectors• Setup, organized, and managed wet lab space, ordered lab supplies, maintained lab inventory, and developed lab protocols
Design Verification Engineer
• Programmed, tested, and validated a GE Healthcare test tool (in C++) that simulates communications between an anesthesia machine and its patient airway measurement unit to facilitate more efficient software testing & design verification• Tested software on anesthesia machines to find software defects within the system • Verified that the anesthesia machines met their system and software design specifications
Undergraduate Research Assistant
• Engineered bacteria to function as biofuel (ethanol/higher alkane-specific) biosensors that produce a fluorescent response proportional to the local biofuel concentration, thereby enabling robust biofuel detection capabilities• Developed a directed evolution assay for improving biosensor response sensitivity and dynamic range• Collaborated on the all-undergraduate UW-Madison international Genetically Engineered Machine (iGEM) Team (2011)
