During my Ph.D. I led a research team that completed the synthesis of the core structure of all mycosamine-containing polyene macrolides, and was part of a team that synthesized several derivatives of the antifungal polyene macrolide amphotericin B. Using these derivatives, we discovered that this family of clinically vital natural products exerts antifungal activity via a mechanism different from that which has been assumed for the past 50 years. These results have important ramifications for the optimization of the therapeutic index of these compounds, the design of new antibiotics which can overcome microbial resistance, and the development of molecular prosthetics as a fundamentally new manifold for the treatment of human disease.
