Some of the tasks carried out were DNA extraction, agarose gel electrophoresis, PCR and confocal microscopy. Other techniques such as manipulation of animal models (mice) and electrophysiology (patch clamp) were performed under supervision. I also performed spinal cord dissection in mice and sample preparation for electrophysiological studies.My work was supervised by Dr. Raúl E. Russo.

Dr. Paruchuri’s lab studies cell signaling aiming to develop novel eicosanoid-based therapies for cancer and cardiovascular diseases. My time in Dr. Paruchuri’s lab consisted of two semesters, fall 2019 and spring 2020. The first semester consisted of cell culture of NHLF cells, performing different treatments in order to see which ones, and to what extent, induced fibrosis by measuring FN and αSMA protein levels. The second semester was centered around the creation of different lentiviral vectors, which would then be used as a vehicle for shRNA delivery of CysLT1R, CysLT2R, P2Y12, GPR17, CREB, and PLKO1 for cellular infection.Some of the tasks I performed were cell seeding, splitting and counting of NHLF and 293FT cells. I also carried out Western Blot, plasmid extraction and the production of lentiviral vectors.Other techniques I was exposed to and either performed them under supervision or observed other members of the lab perform were RNA purification and quantification, cDNA synthesis, qPCR, calcium flux, flow cytometry, ELISA, and working with mice models.

In this internship I contributed to a project based on the cellular mechanism involved in the pathogenesis of Amyotrophic Lateral Sclerosis (ALS).At the Pasteur Institute I mainly analyzed microglia cell cultures and the motor activity of rats with the SOD1G93A mutation. I was also introduced to immunochemistry, as well as confocal and epifluorescence microscopy.My work was supervised by Director Luis Barbeito.

I worked in a project involving a recombinant protein. My tasks revolved around the processes of cloning, expression and purification of this enzyme in different strains of E. Coli.My work was supervised by Dr. Lorena Betancor.
