BTEC658 - Principles in Downstream ProcessingBTEC 658 is a comprehensive study of fundamentals of downstream processing for biochemical products recovery and formulation. Specifically focuses on purification process design and optimization to separate, concentrate and recover a desired product (enzymes, chemicals, pharmaceuticals, novel protein constructs) for its intended use. Integration of innovation in process development, scale-up and facility fit, and commercial process goals under good manufacturing practices is also presented using case studies and publications form reliable sources.Topic areas to be discussed include:(1) learning traditional unit operations, new concepts and emerging technology and their benefits to biochemical product recovery (principles of centrifugation, chromatography, filtration, membrane processes, introduction to continuous processing);(2) assessing the impact of change in unit operations on the process and product quality attributes; (3) examining analytical and process validation issues that are critical to successful manufacturing in large scale a high-purity and high-quality protein; (4) addressing strategy on formulation development and stability; and (5) highlighting validation and regulatory concerns related to downstream process implementation.

Research Assistant
Developed novel methodology for examining residue specific equilibrium unfolding constants of a disease related protein, Aβ. Equilibrium unfolding constants were used to quantify local differences in solvent accessibility of residues suspected to be important in cell association of peptide. MALDI-TOF and ESI-MS coupled with HPLC or LC, together with chemical modification were uses as tools to examine Aβ protein structure. Implemented kinetic analyses along with mathematical modeling and parameter estimation strategies to obtain quantitative results.Explored Aβ-cell membrane and identified key proteins involved in Aβ induced biological activity utlilizing FCS, confocal microscopy and FRET, supported by cell culture and immonocytochemistry assays. Demonstrated relationship between Aβ solvent accessible residues and biological response.
Teacher Assistant (Ta)
Assisted instructor in undergraduate level chemical engineering classes. Held office hours, lectured in recitation sections and graded homework. Designed and tested laboratory experiments for Unit Operations laboratory and trained new TAs in running laboratory experiments.Courses include: Heat and Mass Transfer, Chemical Engineering Analysis and Biochemical Engineering Laboratory.
Grad Summer Intern
This was a summer intership in 2006. I was working in two departments: Natural Product Discovery and Analytical Chemistry.I though that taking 3 months off from my PhD research and gain experience working in a big Pharmaceutical Company was extremely important to understand the "World behind School" here in the USA.
